Peptides are widely found in natural organisms and play a variety of biological functions in human body. Compared with traditional materials, peptides have lower immunogenicity and better biocompatibility, which makes them ideal biological materials. Self-aggregating peptide is a kind of peptide chain that can be assembled by covalent binding of monomers under physiological or pathological conditions. Their advantages include the ability to aggregate by themselves and their low immunogenicity and biocompatibility as peptides. Therefore, self-aggregating peptides have various potential applications in biology and medical field. Since self-aggregating peptide can self-assemble into insoluble particles under certain conditions, wrapping the target protein with self-aggregating peptide can achieve the purpose of protein purification by simple centrifugation. This method is superior to the existing protein purification methods, since the experimental efficiency and cost can be optimized. Moreover, self-aggregating peptides, because of their peptide nature, do not elicit strong immune responses. Thus, self-aggregating peptides can be loaded with a large number of drugs and can be biodegraded into unharmful products which can be smoothly absorbed by the human bodies. The self-aggregated peptide loaded subunit vaccine can successfully induce and enhance immune response without adverse reactions, which is superior to existing immune adjuvants. Self-aggregated peptide also helps keeping the vaccine active for a long time and effectively reduce the cost of vaccine promotion. Select self-aggregating peptides that can be cleaved under pathological conditions to load drugs can help achieve drug targeting. In addition, the self-aggregating peptides loaded with drugs can be stored in specific sites for a long time, so that it does not require repeated dosing, thus improving the efficiency of drug administration. In this paper, the design of peptide self-assembly based solutions and their different biomedical applications are summarized, and their future prospects are prospected.

Effective treatment of cancer has always been a medical field that is constantly being explored all over the world. Currently, the main means of cancer treatment are surgery, chemotherapy and radiotherapy. In recent years, the application of nanomaterials in the field of biomedicine has been highly anticipated. Nanomedicine is safer and more effective than traditional therapies, especially in distinguishing cancer cells from normal tissues. Nanomedicine plays an irreplaceable role in targeted drug controlled release system. Among them, silica nanoparticle-based and functionalized systems with highly controllable size and shape, low toxicity, good biocompatibility, stability in physiological conditions, and stimuli-responsive capping agents have promoted this research. A functional polymer with dual or triple response to silica nanocarriers provides a reliable solution for precise control of release and enhancement of cancer curative effect. This paper focuses on the experimental research of introducing multifunctional stimulus-responsive mesoporous silica nano-drug carriers, and provides some opinions on typical successful drug delivery, which are as follows. (1) Dual-pH sequential release of arsenic trioxide (ATO) and paclitaxel (PTX) was achieved using mesoporous silica nanoparticles coated with polyacrylic acid (PAA) and pH sensitive lipids (PSL); (2) Efficient encapsulation of DOX with HMSN, followed by β-cyclodextrin overlaid on the disulfide bond of glutathione, and finally the surface of the particles was decorated with amantanide-containing citrate functionalized polylysine (PLL(cit)-Ad) for pH/GSH-dual responsive controlled release through subject-object interaction; (3) Atom transfer radical polymerization which is initiated on surface developed based on PDEAEMA-modified hollow mesoporous silica nanoparticles (HMSN) as pH-, reduction-, and light-responsive nanocarriers. Finally, potential problems and development prospects of multifunctional MSNs for biocarrier are summarized.

Sleep is a vital function to all species for reinvigorating the mental and physical senses. During sleep, specific synapses become less active to eliminate unneeded memories as other synapses strengthen to enhance memory consolidation. Healthy quality and longevity of sleep can also regulate moods by ridding fatigue and repairing integrity of the medial prefrontal cortex-amygdala connections. External disturbances affect the quality of sleep, and an individual's routine may play a role in the development of sleeping disorders. By irregulating the circadian rhythm and having consistent negative sleeping habits, sleep disorders such as insomnia, narcolepsy, and sleep apnea may develop. Eight to nine hours of sleep are recommended to both teenagers and adults, if less is received, difficulty in concentration and interpretation of information may be present, as well as physiological effects such as higher blood pressure caused by the disrupted regulation of stress hormones, which if continued, can make the body more prone to serious health conditions.The science of sleep and dreaming comprises a relevant thesis in modern neuroscience research and issues major comprehensions on the study of human consciousness. Dreams are an integral part of sleep. Sleep and dream cycles vary in phenomenology, characteristics, and under circumstances, interpretation of the individual. Various theories such as Sigmun Freud's Psychodynamic theory, the activation synthesis model, and the cognitive development theory have been created and numerous studies are ongoing as the principal motive of dreams has not yet been concluded. Some theories remain heavily physiological as high difficulty of investigations are discontinued by scientists and deemed unverifiable. The research presented here revolves around the neurology of sleep as to inquire into the physiology and investigations on the purpose of dreams. We thus conclude the current review with a call for further research directed towards the primary purpose of sleep and dream.

Due to the inability of immune system to identify certain proteins as self-antigens, rheumatoid arthritis (RA), a chronic, inflammatory, and autoimmune disorder that predominantly affects the joints, causes inflammation in the afflicted body parts. Both hereditary and environmental variables converge to cause RA. Anti-angiogenesis likely serves as a treatment approach as it is crucial to the maintenance of synovitis in RA. sFlt-1 is an angiogenic factor which functions by interacting with VEGF cytokines to compete with VEGF receptor to bind VEGF in order to obstruct cell signaling and hence halt angiogenesis. The absence of the transmembrane and cytoplasmic domains of sFlt-1, an alternatively spliced version of the VEGF receptor-1, ensures that binding will not cause inflammation. Decreased production of new blood vessels results in less joint inflammation and pannus development. That is what the therapy aims to treat so sFlt-1 is appropriate for use in anti-angiogenic therapy. In addition to the pathogenesis of the RA and the workings mechanism of sFlt-1, this article also mentions the detailed manufacturing process of sFlt-1: vector selected with specific HaloTag® tag, transformation by heat shock, transfection in A549 cells, extraction from culture and purification through HaloTag® protein purification and SEC method. Although the entire production process seems seamless, it can become very challenging in practice. Although challenging, the circumstances in which the biological reagents are maintained and cultured cells must be tightly managed. Besides, other social, ethical and economic concerns should also be taken into account. The system used specifically is the vesicular drug delivery system. Additionally, difficult therapeutic testing of sFLit-1is inevitable. sFlt-1 is still of significant research interest and has the potential to develop into a novel commercial treatment, even though there is not enough experimental evidence to demonstrate how successful it is in treating RA as a disease.

Every year, few novel drugs were approved by FDA, getting the chance to go public. Their treatment on different diseases interested the authors to find the reason why a certain drug can apply on the treatment from the initial state to the finished medicines and the efforts scientists took for the therapy of diseases. The Rukobia is a good option approved by FDA in 2020 as a novel HIV-1 treatment drug which can help the treatment for multi-drug resistance patients. With the rapidly increasing HIV patients, developing countries need to control and cure the patients with HIV and the treatment for those multi-drug resistance patients can effectively prolong their lives. Present drug therapy can’t satisfy the necessary of AIDS patients and the usual treatment cocktail therapy bring patients high drug resistance of first line drugs which means the patients will have no excellent alternative treatment after the cocktail therapy, finishing their lives with no hope. Although the HIV can be treated by the specific gene treatment but it can’t be applied on most of patients and the completely curative patients are only 4 until now, as a result, the drug treatment is still important for HIV therapy. Rukobia can’t cure the AIDS because the high replication rate of the HIV can’t be completely killed in the short time so the drug resistance will also produce after a period of treatment but the novel active site make it different from the known antiviral drugs. The authors want to explore the mechanism of Rukobia and the relationship between the drug development and treatment so Rukobia is a good option which applied the new standard and new methods so it has strong reference meaning for this review. In the future, scientists including the authors can apply the mentioned researching method the development and treatment of the new drugs for other diseases and the Rukobia treatment is summarized more clearly in this review.
