Advanced melanoma has now become a severe health threat to both male and female around the globe. With improvements in treatment strategies and diagnosis techniques, prognosis of advanced melanoma patients is becoming better. However, most breakthroughs regarding melanoma treatment are centered around therapies such as immune checkpoint blockade, with less attention being paid on therapeutic cancer vaccines, a type of cancer immunotherapy. In this paper, the mechanism and clinical progress of five types of therapeutic cancer vaccines in treating advanced melanoma are summarized, and an additional focus on the tumor microenvironment of melanoma is included. Although therapeutic cancer vaccines sound promising, a combinational therapy with other immunotherapies, including CAR-T cell therapy and immune checkpoint blockade, is recommended to harvest more clinical benefits for melanoma patients.
